The Excited State Dynamics of a Mutagenic Cytidine Etheno Adduct Investigated by Combining Time-Resolved Spectroscopy and Quantum Mechanical Calculations

Joint femtosecond fluorescence upconversion experiments and theoretical calculations provide a hitherto unattained degree of characterization and understanding of the mutagenic etheno adduct 3,N4-etheno-2'-deoxycytidine (εεdC) excited state relaxation. This endogenously formed lesion is attracting great interest because of its ubiquity in human tissues and its highly mutagenic properties. The εεdC fluorescence is modified with respect to that of the canonical base dC, with a 3-fold increased lifetime and quantum yield at neutral pH. This behavior is amplified upon protonation of the etheno ring (εεdCH+). Quantum mechanical calculations show that the lowest energy state ππππ*1 is responsible for the fluorescence and that the main nonradiative decay pathway to the ground state goes through an ethene-like conical intersection, involving the out-of-plane motion of the C5 and C6 substituents. This conical intersection is lower in energy than the ππππ* state (ππππ*1) minimum, but a sizable energy barrier explains the increase of εεdC and εεdCH+ fluorescence lifetimes with respect to that of dC.

Generalized projection retrieval of dispersion scans for ultrashort pulse characterization

We present a retrieval algorithm based on generalized projections for ultrashort pulse characterization using dispersion scan (d-scan). The new algorithm is tested on several simulated cases and in two different experimental cases in the few-cycle regime. The proposed algorithm is much faster and leads to a drastic reduction of retrieval times, but performs less robust in the retrieval of noisy d-scan traces compared to the standard algorithm.

Spatiotemporal characterization of ultrashort optical vortex pulses

Generation of few-cycle optical vortex pulses is challenging due to the large spectral bandwidths, as most vortex generation techniques are designed for monochromatic light. In this work, we use a spiral phase plate to generate few-cycle optical vortices from an ultrafast titanium:sapphire oscillator, and characterize them in the spatiotemporal domain using a recently introduced technique based on spatially resolved Fourier transform spectrometry. The performance of this simple approach to the generation of optical vortices is analyzed from a wavelength dependent perspective, as well as in the spatiotemporal domain, allowing us to completely characterize ultrashort vortex pulses in space, frequency, and time.

Simultaneous compression and characterization of ultrashort laser pulses using chirped mirrors and glass wedges

We present a simple and robust technique to retrieve the phase of ultrashort laser pulses, based on a chirped mirror and glass wedges compressor. It uses the compression system itself as a diagnostic tool, thereby making unnecessary the use of complementary diagnostic tools. We used this technique to compress and characterize 7.1 fs laser pulses from an ultrafast laser oscillator.
