Efficient Estimation For The Cox Proportional Hazards Cure Model

While analysing time-to-event data, it is possible that a certain fraction of subjects will never experience the event of interest and they are said to be cured. When this feature of survival models is taken into account, the models are commonly referred to as cure models. In the presence of covariates, the conditional survival function of the population can be modelled by using cure model which depends on the probability of being uncured (incidence) and the conditional survival function of the uncured subjects (latency), and a combination of logistic regression and Cox proportional hazards (PH) regression is used to model the incidence and latency respectively. In this paper, we have shown the asymptotic normality of the profile likelihood estimator via asymptotic expansion of the profile likelihood and obtain the explicit form of the variance estimator with an implicit function in the profile likelihood. We have also shown the efficient score function based on projection theory and the profile likelihood score function are equal. Our contribution in this paper is that we have expressed the efficient information matrix as the variance of the profile likelihood score function. A simulation study suggests that the estimated standard errors from bootstrap samples (SMCURE package) and the profile likelihood score function (our approach) are providing similar and comparable results. The numerical result of our proposed method is also shown by using the melanoma data from SMCURE R-package (Cai et al., 2012) and we compare the results with the output obtained from SMCURE package.

On differentiability of implicitly defined function in semi-parametric profile likelihood estimation

In this paper, we study the differentiability of implicitly defined functions which we encounter in the profile likelihood estimation of parameters in semi-parametric models. Scott and Wild (Biometrika 84 (1997) 57-71; J. Statist. Plann. Inference 96 (2001) 3-27) and Murphy and van der Vaart (J. Amer. Statist. Assoc. 95 (2000) 449-485) developed methodologies that can avoid dealing with such implicitly defined functions by parametrizing parameters in the profile likelihood and using an approximate least favorable submodel in semi-parametric models. Our result shows applicability of an alternative approach presented in Hirose (Ann. Inst. Statist. Math. 63 (2011) 1247-1275) which uses the direct expansion of the profile likelihood.

Concentration fluctuations and phase transitions in coupled modulated bilayers

We consider the formation of finite-size domains in lipid bilayers consisting of saturated and hybrid lipids. First, we describe a monolayer model that includes a coupling between a compositional scalar field and a two-dimensional vectorial order-parameter. Such a coupling yields an effective two-dimensional microemulsion free-energy for the lipid monolayer, and its characteristic length of compositional modulations can be considered as the origin of finite-size domains in biological membranes. Next, we consider a coupled bilayer composed of two modulated monolayers, and discuss the static and dynamic properties of concentration fluctuations above the transition temperature. We also investigate the micro-phase separation below the transition temperature, and compare the micro-phase separated structures with statics and dynamics of concentration fluctuations above the transition.

Reparametrization of the least favorable submodel in semi-parametric multisample models

The method of estimation in Scott and Wild (Biometrika 84 (1997) 57--71 and J. Statist. Plann. Inference 96 (2001) 3--27) uses a reparametrization of the profile likelihood that often reduces the computation times dramatically. Showing the efficiency of estimators for this method has been a challenging problem. In this paper, we try to solve the problem by investigating conditions under which the efficient score function and the efficient information matrix can be expressed in terms of the parameters in the reparametrized model.

Coupled Modulated Bilayers

We propose a model addressing the coupling mechanism between two spatially modulated monolayers. We obtain the mean-field phase diagrams of coupled bilayers when the two monolayers have the same preferred modulation wavelength. Various combinations of the monolayer modulated phases are obtained and their relative stability is calculated. Due to the coupling, a spatial modulation in one of the monolayers induces a similar periodic structure in the second one. We have also performed numerical simulations for the case when the two monolayers have different modulation wavelengths. Complex patterns may arise from the frustration between the two incommensurate but annealed structures.
