AlphaFold Accelerates Artificial Intelligence Powered Drug Discovery: Efficient Discovery of a Novel Cyclin-dependent Kinase 20 (CDK20) Small Molecule Inhibitor

The AlphaFold computer program predicted protein structures for the whole human genome, which has been considered as a remarkable breakthrough both in artificial intelligence (AI) application and structural biology. Despite the varying confidence level, these predicted structures still could significantly contribute to structure-based drug design of novel targets, especially the ones with no or limited structural information. In this work, we successfully applied AlphaFold in our end-to-end AI-powered drug discovery engines constituted of a biocomputational platform PandaOmics and a generative chemistry platform Chemistry42, to identify a first-in-class hit molecule of a novel target without an experimental structure starting from target selection towards hit identification in a cost- and time-efficient manner. PandaOmics provided the targets of interest and Chemistry42 generated the molecules based on the AlphaFold predicted structure, and the selected molecules were synthesized and tested in biological assays. Through this approach, we identified a small molecule hit compound for CDK20 with a Kd value of 8.9 +/- 1.6 uM (n = 4) within 30 days from target selection and after only synthesizing 7 compounds. Based on the available data, the second round of AI-powered compound generation was conducted and through which, a more potent hit molecule, ISM042-2 048, was discovered with a Kd value of 210.0 +/- 42.4 nM (n = 2), within 30 days and after synthesizing 6 compounds from the discovery of the first hit ISM042-2-001. To the best of our knowledge, this is the first reported small molecule targeting CDK20 and more importantly, this work is the first demonstration of AlphaFold application in the hit identification process in early drug discovery.

Systolic blood pressure estimation using ECG and PPG in patients undergoing surgery

Background and Objectives: In a significant portion of surgeries, blood pressure (BP) is often measured non-invasively in an intermittent manner. This practice has a risk of missing clinically relevant BP changes between two adjacent intermittent BP measurements. This study proposes a method to non-invasively estimate systolic blood pressure (SBP) with high accuracy in patients undergoing surgery. Methods: Continuous arterial BP, electrocardiography (ECG), and photoplethysmography (PPG) signals were acquired from 29 patients undergoing surgery. After extracting 9 features from the PPG and ECG signals, we dynamically selected features upon each intermittent measurement (every 10 min) of SBP based on feature robustness and the principle of correlation-based feature selection. Finally, multiple linear regression models were built to combine these features to estimate SBP every 30 s. Results: Compared to the reference SBP, the proposed method achieved a mean of difference at 0.08 mmHg, a standard deviation of difference at 7.97 mmHg, and a correlation coefficient at 0.89 (p < 0.001). Conclusions: This study demonstrates the feasibility of non-invasively estimating SBP every 30 s with high accuracy during surgery by using ECG, PPG, and intermittent SBP measurements every 10 min, which meets the standard of the Association for the Advancement of Medical Instrumentation. The proposed method has the potential to enhance BP monitoring in the operating room, improving patient outcomes and experiences.

The Proper Motion of the Pulsar J1124-5916 in the Galactic Supernova Remnant G292.0+1.8

We present the first direct measurement of the proper motion of pulsar J1124-5916 in the young, oxygen-rich supernova remnant G292.0+1.8. Using deep Chandra ACIS-I observations from 2006 and 2016, we measure a positional change of $0.^{\prime\prime}21$ $\pm$ $0.^{\prime\prime}05$ over the $\sim$ 10 year baseline, or $\sim$ $0.^{\prime\prime}02$ yr$^{-1}$. At a distance of 6.2 $\pm$ 0.9 kpc, this corresponds to a kick velocity in the plane of the sky of $\mathrm{612\pm 152\,km \, s^{-1}}$. We compare this direct measurement against the velocity inferred from estimates based on the center of mass of the ejecta. Additionally, we use this new proper motion measurement to compare the motion of the neutron star to the center of expansion of the optically emitting ejecta. We derive an age estimate for the supernova remnant of $\gtrsim$ 2000 years. The high measured kick velocity is in line with recent studies of high proper motion neutron stars in other Galactic supernova remnants, and consistent with a hydrodynamic origin to the neutron star kick.

Human Gender Classification: A Review

Gender contains a wide range of information regarding to the characteristics difference between male and female. Successful gender recognition is essential and critical for many applications in the commercial domains such as applications of human-computer interaction and computer-aided physiological or psychological analysis. Some have proposed various approaches for automatic gender classification using the features derived from human bodies and/or behaviors. First, this paper introduces the challenge and application for gender classification research. Then, the development and framework of gender classification are described. Besides, we compare these state-of-the-art approaches, including vision-based methods, biological information-based method, and social network information-based method, to provide a comprehensive review in the area of gender classification. In mean time, we highlight the strength and discuss the limitation of each method. Finally, this review also discusses several promising applications for the future work.
