High-Resolution Limited-Angle Phase Tomography of Dense Layered Objects Using Deep Neural Networks

We present a Machine Learning-based method for tomographic reconstruction of dense layered objects, with range of projection angles limited to $\pm $10$^\circ$. Whereas previous approaches to phase tomography generally require two steps, first to retrieve phase projections from intensity projections and then perform tomographic reconstruction on the retrieved phase projections, in our work a physics-informed pre-processor followed by a Deep Neural Network (DNN) conduct the three-dimensional reconstruction directly from the intensity projections. We demonstrate this single-step method experimentally in the visible optical domain on a scaled up integrated circuit phantom. We show that even under conditions of highly attenuated photon fluxes a DNN trained only on synthetic data can be used to successfully reconstruct physical samples disjoint from the synthetic training set. Thus, the need of producing a large number of physical examples for training is ameliorated. The method is generally applicable to tomography with electromagnetic or other types of radiation at all bands.

A Learning Approach to Optical Tomography

We describe a method for imaging 3D objects in a tomographic configuration implemented by training an artificial neural network to reproduce the complex amplitude of the experimentally measured scattered light. The network is designed such that the voxel values of the refractive index of the 3D object are the variables that are adapted during the training process. We demonstrate the method experimentally by forming images of the 3D refractive index distribution of cells.

Digital confocal microscopy through a multimode fiber

Acquiring high-contrast optical images deep inside biological tissues is still a challenging problem. Confocal microscopy is an important tool for biomedical imaging since it improves image quality by rejecting background signals. However, it suffers from low sensitivity in deep tissues due to light scattering. Recently, multimode fibers have provided a new paradigm for minimally invasive endoscopic imaging by controlling light propagation through them. Here we introduce a combined imaging technique where confocal images are acquired through a multimode fiber. We achieve this by digitally engineering the excitation wavefront and then applying a virtual digital pinhole on the collected signal. In this way, we are able to acquire images through the fiber with significantly increased contrast. With a fiber of numerical aperture 0.22, we achieve a lateral resolution of 1.5um, and an axial resolution of 12.7um. The point-scanning rate is currently limited by our spatial light modulator (20Hz).
