From WSI-level to Patch-level: Structure Prior Guided Binuclear Cell Fine-grained Detection

Accurately and quickly binuclear cell (BC) detection plays a significant role in predicting the risk of leukemia and other malignant tumors. However, manual microscopy counting is time-consuming and lacks objectivity. Moreover, with the limitation of staining quality and diversity of morphology features in BC microscopy whole slide images (WSIs), traditional image processing approaches are helpless. To overcome this challenge, we propose a two-stage detection method inspired by the structure prior of BC based on deep learning, which cascades to implement BCs coarse detection at the WSI-level and fine-grained classification in patch-level. The coarse detection network is a multi-task detection framework based on circular bounding boxes for cells detection, and central key points for nucleus detection. The circle representation reduces the degrees of freedom, mitigates the effect of surrounding impurities compared to usual rectangular boxes and can be rotation invariant in WSI. Detecting key points in the nucleus can assist network perception and be used for unsupervised color layer segmentation in later fine-grained classification. The fine classification network consists of a background region suppression module based on color layer mask supervision and a key region selection module based on a transformer due to its global modeling capability. Additionally, an unsupervised and unpaired cytoplasm generator network is firstly proposed to expand the long-tailed distribution dataset. Finally, experiments are performed on BC multicenter datasets. The proposed BC fine detection method outperforms other benchmarks in almost all the evaluation criteria, providing clarification and support for tasks such as cancer screenings.

Highly fluorescent copper nanoclusters for sensing and bioimaging

Metal nanoclusters (NCs), typically consisting of a few to tens of metal atoms, bridge the gap between organometallic compounds and crystalline metal nanoparticles. As their size approaches the Fermi wavelength of electrons, metal NCs exhibit discrete energy levels, which in turn results in the emergence of intriguing physical and chemical (or physicochemical) properties, especially strong fluorescence. In the past few decades, dramatic growth has been witnessed in the development of different types of noble metal NCs (mainly AuNCs and AgNCs). However, compared with noble metals, copper is a relatively earth-abundant and cost-effective metal. Theoretical and experimental studies have shown that copper NCs (CuNCs) possess unique catalytic and photoluminescent properties. In this context, CuNCs are emerging as a new class of nontoxic, economic, and effective phosphors and catalysts, drawing significant interest across the life and medical sciences. To highlight these achievements, this review begins by providing an overview of a multitude of factors that play central roles in the fluorescence of CuNCs. Additionally, a critical perspective of how the aggregation of CuNCs can efficiently improve the florescent stability, tunability, and intensity is also discussed. Following, we present representative applications of CuNCs in detection and bioimaging. Finally, we outline current challenges and our perspective on the development of CuNCs.

Chance Constrained Mixed Integer Program: Bilinear and Linear Formulations, and Benders Decomposition

In this paper, we study chance constrained mixed integer program with consideration of recourse decisions and their incurred cost, developed on a finite discrete scenario set. Through studying a non-traditional bilinear mixed integer formulation, we derive its linear counterparts and show that they could be stronger than existing linear formulations. We also develop a variant of Jensen's inequality that extends the one for stochastic program. To solve this challenging problem, we present a variant of Benders decomposition method in bilinear form, which actually provides an easy-to-use algorithm framework for further improvements, along with a few enhancement strategies based on structural properties or Jensen's inequality. Computational study shows that the presented Benders decomposition method, jointly with appropriate enhancement techniques, outperforms a commercial solver by an order of magnitude on solving chance constrained program or detecting its infeasibility.

Y Chromosomes of 40% Chinese Are Descendants of Three Neolithic Super-grandfathers

Demographic change of human populations is one of the central questions for delving into the past of human beings. To identify major population expansions related to male lineages, we sequenced 78 East Asian Y chromosomes at 3.9 Mbp of the non-recombining region (NRY), discovered >4,000 new SNPs, and identified many new clades. The relative divergence dates can be estimated much more precisely using molecular clock. We found that all the Paleolithic divergences were binary; however, three strong star-like Neolithic expansions at ~6 kya (thousand years ago) (assuming a constant substitution rate of 1e-9/bp/year) indicates that ~40% of modern Chinese are patrilineal descendants of only three super-grandfathers at that time. This observation suggests that the main patrilineal expansion in China occurred in the Neolithic Era and might be related to the development of agriculture.
