Latent Class Analysis with Semi-parametric Proportional Hazards Submodel for Time-to-event Data

Latent class analysis (LCA) is a useful tool to investigate the heterogeneity of a disease population with time-to-event data. We propose a new method based on non-parametric maximum likelihood estimator (NPMLE), which facilitates theoretically validated inference procedure for covariate effects and cumulative hazard functions. We assess the proposed method via extensive simulation studies and demonstrate improved predictive performance over standard Cox regression model. We further illustrate the practical utility of the proposed method through an application to a mild cognitive impairment (MCI) cohort dataset.

Quantile regression on inactivity time

The inactivity time, or lost lifespan specifically for mortality data, concerns time from occurrence of an event of interest to the current time point and has recently emerged as a new summary measure for cumulative information inherent in time-to-event data. This summary measure provides several benefits over the traditional methods, including more straightforward interpretation yet less sensitivity to heavy censoring. However, there exists no systematic modeling approach to inferring the quantile inactivity time in the literature. In this paper, we propose a regression method for the quantiles of the inactivity time distribution under right censoring. The consistency and asymptotic normality of the regression parameters are established. To avoid estimation of the probability density function of the inactivity time distribution under censoring, we propose a computationally efficient method for estimating the variance-covariance matrix of the regression coefficient estimates. Simulation results are presented to validate the finite sample properties of the proposed estimators and test statistics. The proposed method is illustrated with a real dataset from a clinical trial on breast cancer.

Globally adaptive quantile regression with ultra-high dimensional data

Quantile regression has become a valuable tool to analyze heterogeneous covaraite-response associations that are often encountered in practice. The development of quantile regression methodology for high-dimensional covariates primarily focuses on examination of model sparsity at a single or multiple quantile levels, which are typically pre-specified ad hoc by the users. The resulting models may be sensitive to the specific choices of the quantile levels, leading to difficulties in interpretation and erosion of confidence in the results. In this article, we propose a new penalization framework for quantile regression in the high-dimensional setting. We employ adaptive L1 penalties, and more importantly, propose a uniform selector of the tuning parameter for a set of quantile levels to avoid some of the potential problems with model selection at individual quantile levels. Our proposed approach achieves consistent shrinkage of regression quantile estimates across a continuous range of quantiles levels, enhancing the flexibility and robustness of the existing penalized quantile regression methods. Our theoretical results include the oracle rate of uniform convergence and weak convergence of the parameter estimators. We also use numerical studies to confirm our theoretical findings and illustrate the practical utility of our proposal
