High-resolution structure of coexisting nanoscopic and microscopic lipid domains

We advanced a previously reported model for the \textit{in situ} scattering data analysis of coexisting lipid domains in free-floating multilamellar vesicles. Based on the scattering density profile of the individual domains we considered in particular modifications due to positionally anticorrelated domains or the presence of cholesterol. The latter aspect can be easily extended to any other lipophilic molecule. This allowed us to retrieve elastic and structural details (bending fluctuations, domain thickness, area per lipid) of coexisting liquid-ordered/liquid-disordered domains, including the partitioning of cholesterol from small-angle x-ray scattering experiments. Moreover, the model also successfully fitted data of multilamellar vesicles exhibiting nanoscopic domains, which displayed unlike coexisting micron-sized domains no clear signal for two lamellar lattices. Our results indicate a previously not observed translation of cholesterol toward the bilayer center of nanoscopic liquid-ordered domains.

Modulation of elasticity and interactions in charged lipid multibilayers: monovalent salt solutions

We have studied the electrostatic screening effect of NaCl solutions on the interactions between anionic lipid bilayers in the fluid lamellar phase using a Poisson-Boltzmann based mean-field approach with constant charge and constant potential limiting charge regulation boundary conditions. The full DLVO potential, including the electrostatic, hydration and van der Waals interactions, was coupled to thermal bending fluctuations of the membranes via a variational Gaussian Ansatz. This allowed us to analyze the coupling between the osmotic pressure and the fluctuation amplitudes and compare them both simultaneously with the measured dependence on the bilayer separation, determined by the small-angle X-ray scattering experiments. High-structural resolution analysis of the scattering data revealed no significant changes of membrane structure as a function of salt concentration. Parsimonious description of our results is consistent with the constant charge limit of the general charge regulation phenomenology, with fully dissociated lipid charge groups, together with a four-fold reduction of the membranes' bending rigidity upon increasing NaCl concentration.

Bending rigidities and interdomain forces in membranes with coexisting lipid domains

In order to precisely quantify the fundamental interactions between heterogeneous lipid membranes with coexisting liquid-ordered (Lo) and liquid-disordered (Ld) domains, we performed detailed osmotic stress SAXS experiments by exploiting the domain alignment in raft-mimicking lipid multibilayers. Performing a Monte Carlo (MC) based analysis allowed us to determine with high reliability the magnitude and functional dependence of interdomain forces concurrently with the bending elasticity moduli. In contrast to previous methodologies, this approach enabled us to consider the entropic undulation repulsions on a fundamental level, without having to take recourse to crudely justified mean-field like additivity assumptions. Our detailed Hamaker coefficient calculations indicated only small differences in the van der Waals attractions of coexisting Lo and Ld phases. In contrast, the repulsive hydration and undulation interactions differed significantly, with the latter dominating the overall repulsions in the Ld phase. Therefore, alignment of like domains in multibilayers appears to originate from both, hydration and undulation repulsions.

Global SAXS data analysis for multilamellar vesicles: Evolution of the scattering density profile (SDP) model

We adapted the highly successful scattering density profile (SDP) model, which is used to jointly analyze small angle x-ray and neutron scattering data from unilamellar vesicles, for use with data from fully hydrated, liquid crystalline multilamellar vesicles (MLVs). By using a genetic algorithm, this new method is capable of providing high resolution structural information, as well as determining bilayer elastic bending fluctuations from standalone x-ray data. Important structural parameters, such as bilayer thickness and area per lipid were determined for a series of saturated and unsaturated lipids, as well as binary mixtures with cholesterol. Results are in good agreement with previously reported SDP data, which used both neutron and x-ray data. The addition of deuterated and non-deuterated MLV neutron data to the analysis improved lipid backbone information, but did not improve, within experimental uncertainties, the structural information regarding bilayer thickness and area per lipid.

Monolayer Spontaneous Curvature of Raft-Forming Membrane Lipids

Monolayer spontaneous curvatures for cholesterol, DOPE, POPE, DOPC, DPPC, DSPC, POPC, SOPC, and egg sphingomyelin were obtained using small-angle X-ray scattering (SAXS) on inverted hexagonal phases (HII). Spontaneous curvatures of bilayer forming lipids were estimated by adding controlled amounts to a HII forming template following previously established protocols. Spontanous curvatures of both phosphatidylethanolamines and cholesterol were found to be at least a factor of two more negative than those of phosphatidylcholines, whose J0 are closer to zero. Interestingly, a significant positive J0 value (+0.1 1/nm) was retrieved for DPPC at 25 °C. We further determined the temperature dependence of the spontaneous curvatures J0(T) in the range from 15 to 55 \degC, resulting in a quite narrow distribution of -1 to -3 * 10^-3 1/nm°C for most investigated lipids. The data allowed us to estimate the monolayer spontaneous curvatures of ternary lipid mixtures showing liquid ordered / liquid disordered phase coexistence. We report spontaneous curvature phase diagrams for DSPC/DOPC/Chol, DPPC/DOPC/Chol and SM/POPC/Chol and discuss effects on protein insertion and line tension.

A defect mediated lamellar to isotropic transition of amphiphile bilayers

We report the observation of a novel isotropic phase of amphiphile bilayers in a mixed system consisting of the ionic surfactant, sodium docecylsulphate (SDS), and the organic salt p-toludine hydrochloride (PTHC). This system forms a collapsed lamellar ($L_α$) phase over a wide range of water content, which transforms into an isotropic phase on heating. This transition is not observed in samples without excess water, where the $L_α$ phase is stable at higher temperatures. Our observations indicate that the $L_α$ - isotropic transition is driven by the unbinding of edge dislocation loops and that the isotropic phase in the present attraction-dominated system is the analogue of the sponge phase usually seen in amphiphile systems dominated by interbilayer steric repulsion.
